Product Description/Introduction
This product is RNase Inhibitor (Ribonuclease Inhibitor), namely ribonuclease inhibitor, which is A porcine derived ribonuclease inhibitor recombinant expressed by E.coli. It binds to RNase A, B and C in a non-competitive manner in a ratio of 1:1.Thus inhibiting the activity of these RNases and protecting RNA from degradation. This ribonuclease inhibitor has been shown to inhibit RNase in a variety of tissues and cells, but not RNase 1, H, T1, S1 Nuclease and RNase from Aspergillus..The performance of porcine and human ribonuclease inhibitors is very similar. The inhibition activity is maintained by at least 1 mM DTT in the reaction system. The effective reaction pH is 5.0-8.0, and the optimal reaction pH is 7.0-8.0.
Uses : cDNA synthesis, in vitro RNA transcription, in vitro protein synthesis, etc.
Source : porcine ribonuclease, recombinant expression in Escherichia coli.
Enzyme activity definition : The amount of enzyme capable of inhibiting 50 % activity of 5 ng RNase A was defined as an enzyme activity unit.
Purity and concentration : SDS-PAGE detection purity ≥ 95 %, 40 U / μL.
Inactivation or inhibition : complete inactivation at 75℃ for 10 min. Protein denaturants such as SDS and urea can inhibit the inhibition of RNase Inhibitor on RNase.
Enzyme storage buffer : 20 mM Tris-HCl, 50 mM NaCl, 5 mM DTT, 50 % ( v / v ) glycerol, pH 7.5.

The total RNA of 1 μg Hela cells, 10 ng RNase A and different amounts of RNase Inhibitor were treated at room temperature for 10 min and analyzed by electrophoresis with 1% agarose gel. As can be seen from the figure, RNase Inhibitor can effectively inhibit RNase A’s degradation of RNA.

Assay Protocol / Procedures
1. For common reaction systems ( cDNA synthesis, in vitro transcription, in vitro protein synthesis, etc. ), in order to protect RNA from degradation by RNase in the system, the recommended final concentration of RNase Inhibitor is 1-2 U / μL.

1. Enzyme products should be placed in an ice box or on ice when used. It is recommended to use them after packaging, and they should be stored at − 20 °C immediately after use.
