Operating Instructions:

  1. Prepare your cell or tissue samples according to the specific requirements of your experiment.
  2. Prepare the BCECF, AM Working Solution: Follow the instructions provided in the product manual to prepare the BCECF, AM Working Solution.
  3. Staining and Labeling:

    3.1. Remove the existing cell culture medium and wash the cells or tissue samples with PBS or an appropriate buffer.

    3.2. Discard the wash buffer and add the prepared BCECF, AM Working Solution. Incubate the samples in the dark at room temperature or 37°C for the designated time.

    3.3. After the incubation period, wash the samples with PBS or buffer to remove excess probe.

  4. Fluorescence Detection: Employ suitable instruments such as fluorescence microscopes, microplate readers, flow cytometers, etc., to quantify changes in intracellular pH by monitoring alterations in the fluorescence intensity of BCECF, AM.
