Description
This product is a safe and harmless buffer for rapid wet membrane transfer in Western Blot, which can efficiently and quickly transfer proteins to the blotting membrane. Without methanol, the film transfer process can be completed within 15-30 min.

Rapid membrane transfer without ice bath: With rapid membrane transfer buffer, proteins on polyacrylamide gels can be transferred to imprinted membranes such as PVDF membrane or nitrate fiber membrane (NC membrane) in a constant flow membrane of 400 mA for 15-30 min, and the heat production is lower than that of conventional membrane transfer buffer.

Assay Protocol
1. Each bag of ice-bath free fast film transfer buffer powder is dissolved in 800 mL pure water, and 200 mL absolute ethanol is added to fully dissolve and mix (it is properly prepared according to experimental requirements);;
2. Complete the sandwich structure required for transfer printing, from the negative electrode (black side) to the positive electrode (red side) in the order of sponge, filter paper, gel, blotting film, filter paper, sponge, and then transfer to the transfer groove, set a constant current of 400 mA, film transfer time of 15-30 min (this condition is for 1.0mm thick gel, such as 0.75mm gel thickness, Appropriately reduce the film transfer time; If the gel thickness is 1.5 mm, extend the transmembrane time appropriately) to complete the protein transmembrane.
Note:
1. This product is suitable for the membrane transfer of PAGE gel system of Tris-Gly, Bis-Tris and other buffer systems.
2. PVDF membrane should be infiltrated with absolute ethanol for about 30 s before use.
3. The transfer current is recommended to be set at a constant current of 400 mA. Generally, proteins below 20 kDa are recommended to be transferred for 5-15 min; 20-150 kDa protein is recommended to transmembrane for 15-25 min; Proteins above 150 kDa are recommended to transmembrane for 25-35 min.
4. Ice bath is not required for film transfer within 30 min. Ice bath is recommended for film transfer over 30 min.
5. Due to the different brand and model of the power supply used for the film, on the one hand, the highest current is different, on the other hand, the power will increase the difference; When the constant current of some power supplies is set to 400 mA, overload protection occurs when the maximum power is exceeded. In this case, you need to lower the current properly or choose a power supply with higher power.
6. The temperature of the transfer buffer, gel parameters (such as thickness, acrylamide concentration, ion concentration), filter paper thickness, volume of transfer buffer and times of use of transfer buffer may affect the current and voltage; In case of overload, please reduce the current and prolong the film turning time.
7. The normal color of this product is colorless and transparent. If light brown or yellowish brown appears, it should be discarded in time.
