Product Name: Glasgow’s MEM (GMEM), Powder

Product Code: G4558P-1L (1 L), G4558P-10L (10 L), G4558P-50L (50 L)

Product Components: This product is a powdered Glasgow’s MEM (GMEM) medium containing 2.0 mM L-alanyl-L-glutamine and phenol red indicator. It does not contain sodium pyruvate or HEPES. Sodium bicarbonate needs to be added separately before use.

Storage and Expiry: This product is transported at room temperature and should be stored in a dry, light-protected area at 2-8°C. The shelf life is three years.

Usage Instructions:

Preparation Steps before Mixing:

  1. Prepare a clean container with a volume close to the final volume.
  2. Prepare distilled water or cell culture-grade water (recommended G4701) at room temperature (15-30°C).
  3. Prepare 1 N NaOH and 1 N HCl.

Steps to Prepare Solution from Powder:

  1. Inject 95% of the final volume of distilled water into the clean container. Take one bottle of this product (for preparing 1 L medium) or weigh the powder proportionally and dissolve it in the water. Note that the powder will dissolve quickly after the breakdown in water. Rapid stirring may cause clumping of the powder and the formation of a water film on the surface, hindering dissolution. Therefore, high-speed stirring is unnecessary after adding the powder to water.
  2. After the powder is dissolved and the solution is clear, add sodium bicarbonate according to a dosage of 2.75 g/L and stir to dissolve. If preparing a different volume, add the corresponding amount of sodium bicarbonate.
  3. Adjust the pH to 0.2-0.3 units below the working pH (recommended working pH 6.7-7.1) using 1 N NaOH and 1 N HCl. The pH will increase by 0.1-0.3 units after filtration.
  4. Add water to reach the final volume. Keep the solution sealed before filtering.
  5. Use positive pressure to filter through a 0.2 μm membrane into a sterile container (observe aseptic procedures).
  6. After filtration, take a small amount of the liquid for aseptic inspection before use. Store the prepared liquid medium at 2-8°C, with a shelf life of one year.

Notes:

  1. The powder is hygroscopic, so seal it promptly after use.
  2. Use proper protective measures when handling the powder to avoid inhalation.
  3. Cover the container holding the medium during stirring and filtration to prevent excessive agitation. Stop stirring after the powder dissolves.
