Description

Product Composition

This product is a powdered RPMI-1640 medium containing 2.05 mM L-alanyl-L-glutamine and a phenol red indicator. It does not contain sodium pyruvate or HEPES. Sodium bicarbonate needs to be added separately when using.

This product is shipped at room temperature and should be stored in a dry place at 2-8°C, protected from light. It has a shelf life of three years.

Instructions for Use

l Preparatory Work before Preparation

1. Prepare a clean container with a volume close to the final volume.
2. Prepare distilled water or cell-grade water (recommended G4701) at room temperature (15-30°C).
3. Prepare 1 N NaOH and 1 N HCl.

l Steps for Preparing Solution from Powder:

1. Inject 95% of the final volume of distilled water into the prepared clean container. Take one bottle of this product (sufficient to prepare 1 L of medium) or weigh the powder proportionally and dissolve it in the water. Note that the powder will dissolve rapidly after disintegration in water. Vigorous stirring may cause clumping of the powder and the formation of a water film on the surface, which is not conducive to dissolution. Therefore, there is no need for high-speed stirring after adding the powder to water.
2. After the powder is dissolved and the solution becomes clear, add sodium bicarbonate at a concentration of 2.0 g/L and stir to dissolve. If preparing a different volume, add an appropriate amount of sodium bicarbonate corresponding to the volume.
3. Adjust the pH to be lower than the working pH by 0.2-0.3 units using 1 N NaOH and 1 N HCl (recommended working pH 7.0-7.4). The pH of the solution will increase by 0.1-0.3 units after filtration.
4. Add water to reach the final volume. Keep the solution sealed before filtration.
5. Filter the solution under positive pressure through a 0.2μm filter membrane into a sterile container (observe aseptic techniques).
6. After filtration, take a small amount of the liquid for aseptic testing and use it only when it passes the test. The prepared liquid medium can be stored at 2-8°C with a shelf life of one year.

Precautions

1. The powder is hygroscopic, so seal it promptly after use.
2. Take necessary precautions when handling the powder to avoid inhalation.
3. Keep the container covered during stirring and filtration to prevent excessive stirring. Stop stirring once the powder is dissolved.
