Product Composition

This product is a powdered RPMI-1640 medium, containing 2.05 mM L-Proline-L-Glutamine, without the indicator Phenol Red, without Sodium Pyruvate, without HEPES. Sodium bicarbonate needs to be added separately when in use.

This product is shipped at room temperature and should be stored in a dry, light-protected place at 2-8°C. The shelf life is three years.

Instructions for Use

Preparation Before Reconstitution

  1. Prepare a clean container with a volume close to the final volume.
  2. Prepare distilled water or cell culture-grade injection water at room temperature (15-30°C) (recommended: G4701).
  3. Prepare 1 N NaOH and 1 N HCl.

Steps for Reconstitution Using Dry Powder:

  1. Inject 95% of the final volume of distilled water into the clean container. Take 1 bottle of this product (for preparing 1 L of medium) or weigh the powder proportionally and dissolve it in the water. Note that the powder dissolves rapidly after disintegration in water. Excessive agitation might cause clumping of the powder and the formation of a water film on the surface, hindering dissolution. Therefore, high-speed agitation is not necessary after adding the powder to water.
  2. After the powder has dissolved and the solution is clear, add sodium bicarbonate to achieve a concentration of 2.0 g/L. Stir to dissolve. If preparing a different volume, add the appropriate amount of sodium bicarbonate according to the corresponding volume.
  3. Adjust the pH to be 0.2-0.3 units lower than the working pH using 1 N NaOH and 1 N HCl (recommended working pH: 7.0-7.4). The pH may increase by 0.1-0.3 units after filtration.
  4. Bring the solution to the final volume by adding water. Keep the solution sealed before filtration.
  5. Use positive pressure to filter the solution through a 0.2μm filter membrane into a sterile container (ensure aseptic conditions).
  6. After filtration, take a small amount of the liquid for aseptic testing. Once it passes, use the solution. The prepared liquid medium can be stored at 2-8°C with a shelf life of 1 year.

Precautions

  1. The dry powder is hygroscopic, so seal it promptly after use.
  2. Use proper protective measures when handling dry powder to prevent inhalation.
  3. Keep the container with the culture medium covered during stirring and filtration processes to prevent excessive agitation. Stop stirring after the dry powder has dissolved.
