Description

Product Ingredients

This product is a powder form of RPMI-1640 medium containing 2.05 mM L-alanyl-L-glutamine and phenol red indicator. It does not contain sodium pyruvate or HEPES. Sodium bicarbonate needs to be added separately when using.

The product is shipped at room temperature and should be stored in a dry, light-protected place at 2-8°C. It has a shelf life of three years.

Instructions for Use

l Preparatory Work Before Preparation

1. Prepare a clean container with a volume close to the final volume.
2. Prepare room temperature (15-30°C) distilled water or cell-grade water for injection (recommended: G4701).
3. Prepare 1 N NaOH and 1 N HCl.

l Steps for Reconstitution with Powder

1. Inject 95% of the final volume of distilled water into the prepared clean container. Take one bottle of this product (for preparing 1 L of medium) or weigh the powder in the appropriate proportion and add it to the water for dissolution. Note that the powder dissolves rapidly after it is dispersed in water. High-speed agitation may cause the powder to agglomerate and form a water film on the surface, which hinders dissolution. Therefore, high-speed stirring is not necessary after adding the powder to the water.
2. After the powder has dissolved and the solution is clear, add sodium bicarbonate to achieve a concentration of 2.0 g/L. Stir until dissolved. If preparing a different volume, add the corresponding amount of sodium bicarbonate according to the volume.
3. Adjust the pH to 0.2-0.3 units below the working pH (recommended working pH 7.0-7.4) using 1 N NaOH and 1 N HCl. The pH of the solution may increase by 0.1-0.3 units after filtration.
4. Add water to reach the final volume. The solution should be sealed before filtration.
5. Filter the solution under positive pressure using a 0.2 μm filter membrane into a sterile container (observe sterile procedures).
6. After filtration, take a small amount of the liquid for sterile inspection. Use it after confirming its sterility. The prepared liquid medium should be stored at 2-8°C and has a shelf life of 1 year.

Precautions

1. The powder is hygroscopic, so seal it promptly after use.
2. Use protective measures when handling the powder to prevent inhalation.
3. Cover the container holding the medium during stirring and filtration to prevent excessive agitation. Stop stirring once the powder is dissolved.
