The Fluo-4 AM Calcium Ion Fluorescence Assay Kit is designed for the detection of intracellular calcium ion (Ca²⁺) levels in live cells using the fluorescent probe Fluo-4 AM. Operating Instructions:
- Prepare the cells or tissue samples as needed for your experiment.
- Prepare the Fluo-4 AM Working Solution: Follow the instructions provided in the product manual to prepare the Fluo-4 AM Working Solution.
- Staining and Labeling:3.1. Remove the original cell culture medium and wash the cells or tissue samples with PBS or appropriate buffer.3.2. Remove the wash buffer and add the prepared Fluo-4 AM Working Solution. Incubate in the dark at room temperature or 37°C for a specified time.
3.3. After the incubation period, wash the samples with PBS or buffer to remove excess probe.
- Fluorescence Detection: Use appropriate instruments such as fluorescence microscopes, microplate readers, flow cytometers, etc., to detect changes in intracellular calcium ion levels through the increased fluorescence signal of Fluo-4 AM.
